Test Principle:

 

The cells are being loaded with the fluorescence indicator Fura-2 AM. Cellspecific esterasis cut the ester bond and activate the indicator to bind free cytosolic Ca 2+. As Fura-2 is a dual excitation indicator (Ex 340nm and 380nm; Em 510nm) the ratio of emission signals can be used for the determination of the [Ca 2+]-concentration.

Test Principle:

The assay is based on the transformation of colourless or weakly dyed tetrazolium salts into intensively coloured formazan derivatives.This reduction requires active mitochondrions which become inactive within minutes after cell lysis. The change in coulor correlates directly with the cell quantity.

Trypsin Inhibition Test

Test Principle:


The assay is based on the trypsin-catalysed breakdown of N-Benzoyl-DL-arginin-p-nitroanild to p-Nitroaniline.
As the inhibition activity correlates with the amount of  p-Nitroaniline produced, the measured concentration (UV-detection at 405nm) allows the classification of the potential drug agent.
